Reaction Cleanup with Zymo Columns

• add 500 µl DNA Binding buffer to coupling reaction
• Place Zymo column in 2 ml collection tube and load sample onto column
• Spin sample through the column at 5000 RPM
• discard the flow through
• apply 200 µl wash buffer to the column and spin at top speed for 5-10 seconds
• apply 200 µl wash buffer and spin for 30 seconds
• move Zymo column to 1.5 ml tube
• apply 8 µl H2O or TE* and wait for 30 seconds
• spin the sample at top speed to elute, 30 seconds
• apply another 8 µl H2O and wait for 30 seconds
• spin through at top speed, 1 minute
